Seller not responding to offer

So after a little marketing, and being contacted directly by sellers, I made two unofficial (starting point) offers last week via email.
So far, neither seller has even acknowledged receiving the email, let alone saying "yes" or "no".

How long before I can assume the offer wasn't taking seriously, and that they have no intention of responding? Should I expect an immediate response, or does it sometimes take several days?

There are no brokers in between our negotiation.

A bona fide offer needs to be made in writing, not by an e-mail, in good contract form. Follow up with a formal offer. Put a dead line or expiration date on all offers, otherwise they can come back a week later and accept when you have made another offer. 72 hrs is plenty of time.

It's also good to speak to sellers if you can to get a meeting of the minds before going to much trouble, especially if you're making low ball offers. :)
