When is the best time of the day to post discussions in order to get the most responses? 

Living in the US with 4 different time zones makes it a bit more difficult to pinpoint but just after a ballpark.

Also weekdays vs weekends?

And of course the discussion needs to be worthy of a response in the first place...

I don't think there is any one best time . . . I think it does have to do with post title and other factors.  That said, you will certainly see slowdowns on holidays, dinner time and in the middle of the night.
